Requires a self-motivated person to work semi-independently with moderate oversight and assistance provided by the supervisor. The primary work is directed at the understanding of the function of T lymphocytes antigen in normal immunology and pathological states. This would include, but not be limited to, a project examining the immune response to infection with Mycobacterium tuberculosis. Performs a variety of routine and non?routine experiments, develops modification of existing techniques, as appropriate, and may identify new techniques. As a member of the research team, gives input into the development and design of experiments, collects, organizes, and interprets data. The position is for 1-2 years only.1.Work includes participation with a group of investigators characterizing the cellular and molecular nature of the T lymphocyte response in animal models of tuberculosis and other infectious diseases. This will involve maintaining breeding colonies of mice, performing infections, administering test substances such as antibodies and cytokines to infected animals, collecting and analyzing data on the health of the infected animals.2.Perform necropsies on infected animals and process the tissues for histological and microbiological analysis.3.Isolate human lymphocytes from the blood of patients or normal volunteers.4.Cellular immunology experiments will include T cell purification, T cell cloning, proliferation experiments, lymphokine secretion experiments, and cytotoxicity studies. This includes maintaining long-term cultures of cell lines and requires aseptic cell culture technique.5.Analyze lymphocytes by flow cytometry, antibody purification and conjugation using protocols already established in the laboratory.6.Performs other duties and responsibilities as directed.7.The applicant will be trained to perform any techniques not currently known, including instruction in the use of personal protective equipment and safe handling of infectious pathogens. The technician will be directed by the supervisor, but after sufficient training, is expected to be able to work independently on the research project with minimal supervision.
